Immunoproteomic Technique Reveals FlaA is an Antigenic Protein Strongly React with Derived Serum from Campylobacter Jejuni Infected Chicken

Abstract
Campylobacter jejuni is a Gram-negative bacterium leading to campylobacteriosis when consumes the contaminated chicken meat with this bacterium. The symptoms of this disease are severe diarrhea, nausea, and abdominal pain and eventually become Guillain-Barre syndrome. The objective of this research was to analyze the antigens of C. jejuni for vaccine development in order to prevent and control C. jejuni infection in chicken. Here, we analyzed proteins from C. jejuni NCTC11168 by using Two-dimensional Electrophoresis(2-DE) and immuno blot with serum of C. jejuni infected chicken. The result showed that 2-DE could separate proteins according to different pI and size of the proteins. After using immuno blot to detect immunogenic proteins with serum from C. jejuni infected chicken, it revealed that three proteins from C. jejuni NCTC11168 strongly reacted with serum from chicken infected with C. jejuni NCTC11168. When performing LCnanoESI- MS/MS for analysis and identifying the type of the proteins, it revealed that they were FlaA, CadF and OprF respectively. We also successfully expressed recombinant FlaA in E.coli strain BL21 and Western blots result showed that recombinant FlaA strongly recognized by infected chicken serum. This recombinant FlaA will be further tested the efficacy as the vaccine in chicken. Keywords - Campylobacter jejuni, 2-DE , protein expression, immnodetection
