Effect of Mesenchymal Stem Cellon VEP ın DIABETIC Rat

Abstract
Stem cell application is an inovation in biomedical field.Mesenchymal Stem Cell (MSC) represent a great promise in regenerative medicine and the treatment are becoming increasingly important in retinal disease. Today, there is no standardization for this therapy. Degenerative retinal diseases are among the main causes of irreversible vision loss due to degeneration of retinal ganglion cells. Visual Evoked Potential (VEP) help to detect the early changes in of neuronal activity from the retina to the occipital cortex. The purpose of this study was to test the possible therapeutic role of bone marrow mesenchymal stem cell (BMSC) on dejenerative retinal states caused by early diabetes comparing with blood glucose level and intraocular pressure. Thirty two Spraque Dawley rats were used their age 2-3 months, weight 150-350gr. All groups were generated randomly. Control (6), Diabetic (10), BMSC (6) and Diabetic+ BMSC (10). Study were performed for 26 days. Diabetic rat model was generated with a single dose of streptozotosin (60mg/kg). MSC injection (1x10⁶/ 4μl) were done to the right eye twice then after 48 hours and left eye was used as control. VEP were recorded before and after the injection followed by 24 and 48 hours for detecting MSC effect on diabetic disorders in early stages. Control and test groups’s weight, body temperature, blood glucose values, intraocular pressure levels were compared. P100 amplitudes of 1-12 Hertz light stimulus and dominant frequence, mean value, minimum and maximum values of 100 ms were compared in groups after and before injection. Control right and left eye’s P100 amplitudes were not found statistically significant. Diabet left eye’s amplitude was decreased after 12 Hz stimulation. Although amplitude was increased after 24 h. from first injection and decreased after 48 h. from second injection in right eyes. Although it was not found statistically significant to 1 and 12 Hz stimulation, amplitude was increased after 24 h. from second injection to 1 Hz stimulation in diabetic rats’ right eyes. When different frequencies of light stimuli creating a meaningful difference (amplitude) in the left eye in diabetic rats, significant increase was observed in both eye after BMSC applications. As a result it was determined that BMSC transplantation for the purpose of improving the retinal degeneration by diabetes cause the increasing in p100 values due to different stimulus characteristics, diabetes caused a decrease in visual messages. It was thougth that BMSC applications should be use in vast area for the aim of retinal regeneration.
